Triptorelin Single-Shot PCT: Tony Huge’s HPTA Restart Protocol

Table of Contents

Most guys mess up post-cycle therapy. They take their last shot, swallow a Nolva, drink a beer, and pray their balls wake up. They won’t — not for months. And in that gap their bloodwork goes sideways: estrogen crashes, mood drops, libido vanishes, half the muscle they worked for evaporates because cortisol is now running the show.

I’ve run more cycles, audits, and recoveries than I care to count, and I’ve watched guys do “PCT” with everything from clomid pyramids to triple-stacked aromatase inhibitors. The single most underrated tool in the recovery kit is a peptide most lifters have never heard of: triptorelin. One injection, properly timed, can restart the HPTA in weeks instead of months. That’s not hype — that’s the actual mechanism, and it’s been used in clinical endocrinology for decades. The bodybuilding world is just slow on the uptake.

What Triptorelin Actually Is (And Why It Restarts You)

Triptorelin is a synthetic GnRH agonist — gonadotropin-releasing hormone. In your hypothalamus, GnRH is the master signal that tells the pituitary to release LH and FSH, which then tell the testes to produce testosterone and sperm. When you run exogenous androgens (test, SARMs, designer compounds), that whole chain shuts down via negative feedback. Your hypothalamus stops releasing GnRH because it sees high androgen levels and assumes the job is done.

Triptorelin walks into that shut-down system and slams the GnRH receptor with a long-acting analog. The pituitary fires a massive LH and FSH pulse — bigger than anything clomid or HCG can provoke — and the Leydig cells inside the testes get a signal so strong they can’t ignore it. Endogenous testosterone production restarts.

That’s the mechanism. The reason single-shot protocols exist is that triptorelin’s half-life and pituitary engagement are long enough that one well-timed dose can do what eight weeks of SERMs do. Doctors use it in IVF protocols and pediatric endocrinology. Bodybuilders started using it sub-rosa in the late 2000s after a few European labs proved the single-shot recovery model in published case series.

My Personal Triptorelin Protocol (After A Heavy Cycle)

Let me be transparent about how I actually use this — not the textbook version, the real-world version.

After any cycle that suppressed me hard — typically anything over 600mg/week of test for more than 12 weeks, or any prolonged SARM run with YK-11, LGD-4033, or RAD-140 — here’s what I do:

  • Day 1 of PCT (after compounds clear): Single subcutaneous injection of triptorelin acetate, 100mcg.
  • Days 1–14: No SERMs. The pituitary is firing — let it work. Adding clomid here just confuses the signal.
  • Day 14 onward: Pull bloodwork. Total test, free test, LH, FSH, estradiol, prolactin, SHBG. (Same panel I describe in my bloodwork guide for TRT patients — get the full panel, not just total T.)
  • If LH and FSH are firing but T is sluggish: add 25mg clomid EOD for 4 weeks.
  • If estradiol is climbing: low-dose aromasin, 6.25mg twice a week. Don’t crush it — see my estrogen management protocol for why over-suppression of E2 is worse than slightly elevated E2.

That’s it. One shot, 14 days of patience, then bloodwork-driven adjustments. I’ve done this protocol after at least eight major cycles in the last decade and the recovery curve is dramatically faster than the SERM-only PCT I used to run.

Real-World Bloodwork: What “Recovery” Looks Like On Triptorelin

The numbers I’ve seen on myself and on guys I’ve coached through this protocol look something like this. Pre-cycle baseline test: 700–900 ng/dL. End of a 16-week heavy cycle: total test reads inflated from exogenous (~1800+), endogenous is essentially zero, LH and FSH are floored.

Three weeks after triptorelin shot, with compounds cleared:

  • LH: usually back into mid-range (4–8 mIU/mL)
  • FSH: similar range, often slightly higher
  • Total test: typically 400–600 ng/dL by week 3, climbing back to baseline by week 6–8
  • Free test: tracks total but watch SHBG — sometimes elevated for the first month
  • Prolactin: occasionally bumps up briefly, then settles

Compare that to clomid-only PCT, where most guys are still chasing 250 ng/dL at week 4 and feeling like garbage. The single-shot triptorelin protocol skips the worst of that low-T window. That alone is why I keep coming back to it.

Where People Get Triptorelin Wrong

Three mistakes I see constantly:

1. Pinning it too early. If exogenous compounds are still active, the pituitary signal won’t translate to Leydig cell activation because the testes are still desensitized. Wait until your last long ester is fully metabolized. Test cypionate: roughly 18–21 days. Test enanthate: 14–18 days. SARMs: 7–10 days for most short-half-life compounds. Don’t rush it.

2. Stacking SERMs immediately. Clomid and tamoxifen also stimulate the HPTA, but they do it through estrogen receptor antagonism at the hypothalamus. Adding them on top of an active triptorelin shot can create a confused signaling environment and sometimes blunts the LH surge. Let triptorelin do its job for two weeks first.

3. Skipping the bloodwork. This is the same mistake guys make on cycle. You can’t manage what you don’t measure. Pull labs at week 2, week 4, and week 8. Adjust based on numbers, not feel — feel lags numbers by weeks.

Stacking Triptorelin With Recovery Peptides

Triptorelin restarts the hormonal axis, but it doesn’t fix the soft tissue damage, joint wear, or systemic inflammation a heavy cycle leaves behind. That’s where I layer in healing peptides during the same recovery window.

The standard recovery stack I run alongside a triptorelin PCT:

  • BPC-157: 250mcg twice daily, subQ near affected tissue. Joint and gut healing. See my full BPC-157 + TB-500 synergy protocol.
  • TB-500: 2mg twice weekly. Systemic inflammation, vascular repair.
  • Ipamorelin or CJC-1295: Low-dose nightly, supports natural GH pulse during the low-T window when sleep quality usually tanks. My CJC-1295/Ipamorelin stack guide covers dosing.
  • TUDCA: 500mg/day for liver support — doubly important if you ran any 17aa orals. My TUDCA protocol is here.

This stack lets the body heal while the HPTA is restarting. Eight weeks in, you come out the other side with bloodwork in range, joints repaired, and gains preserved.

The FDA, The Gray Market, And Practical Sourcing

Triptorelin in the U.S. is FDA-approved for prostate cancer treatment under the brand name Trelstar — 22.5mg long-acting depot, prescribed only by oncologists. The single-shot bodybuilding protocol uses 100mcg, which is roughly two hundred and twenty times less than the medical dose. That dosage gap is why the compound has remained niche: nobody is writing a prescription for 100mcg of Trelstar to a 32-year-old lifter.

The compound exists in the research peptide market where it’s sold as triptorelin acetate. Quality varies wildly. If you’re going this route, the same rules apply that I cover in my FDA peptide regulation breakdown — third-party tested, reputable supplier, lyophilized vial reconstituted with bacteriostatic water, stored cold. Don’t buy from Instagram.

The political backdrop matters here too. The Kennedy administration’s push to expand peptide access has shifted the regulatory conversation, and compounding pharmacies are slowly opening doors that were locked five years ago. We’re not at the point where you can ask your GP for a triptorelin recovery script, but the trajectory is moving.

Side Effects: What Actually Happens

The first 48 hours after a triptorelin shot can feel weird because the LH surge is large. Some guys report:

  • Mild headache, lasts a day or two
  • Brief libido bump (sometimes uncomfortable for a week, then normalizes)
  • Slight injection site irritation
  • Occasional brief mood lift from the testosterone restart

Long-term side effects from a single 100mcg dose are minimal in the literature. The medical depot doses cause issues because they sustain GnRH receptor downregulation for months — that’s the desired effect for cancer treatment but the opposite of what we want for recovery. The single-shot recovery dose creates a brief pituitary stimulation, not chronic suppression.

Who Shouldn’t Use This

If you’ve been on TRT for years and you’re not trying to come off — triptorelin is irrelevant. You’re on permanent exogenous, no recovery needed. If you’ve never run a real cycle and you’re just doing TRT-doses of test for life optimization, your HPTA is suppressed but you don’t need a restart.

If you have any history of pituitary tumors, adolescent precocious puberty treatment, or active prostate disease — talk to a doctor first. The molecule has more interactions in those populations than the average healthy lifter is aware of.

The Bottom Line From Pattaya

I’ve recovered from cycles in three different decades using three different generations of PCT protocols. SERMs alone are slow. HCG works but blunts the long-term restart and adds estrogen pressure. Triptorelin single-shot, properly timed, with a clean recovery stack underneath it, is the cleanest, fastest restart I’ve personally run.

It’s not magic. You still need bloodwork. You still need to wait the right number of days. You still need to manage estrogen and prolactin and sleep. But the difference between “month four and still in the dumps” and “back to baseline by week eight” is the kind of edge that matters when you’re trying to keep cycling productively over a 20-year career.

Run your PCT smart. Measure everything. And if you’re going to use triptorelin, use it once, time it correctly, and then trust the mechanism to do its work.

Frequently Asked Questions

How does triptorelin work for PCT recovery compared to traditional methods?

Triptorelin is a GnRH agonist that directly stimulates the pituitary to restart natural testosterone production faster than SERMs alone. Unlike nolvadex which blocks estrogen receptors, triptorelin jumpstarts your HPTA axis at the source, reducing the recovery window from months to weeks and preventing the hormonal crash that causes muscle loss and mood crashes.

What is the optimal triptorelin dosage and timing for post-cycle therapy?

Most protocols use a single 42mg depot injection administered 7-10 days after the final steroid shot. This timing allows remaining exogenous hormones to clear while the triptorelin begins signaling HPTA recovery. The extended-release formulation provides sustained GnRH stimulation over weeks, making it more effective than multiple smaller doses for rapid axis restart.

Can you take triptorelin with SERMs like nolvadex during PCT?

Yes, combining triptorelin with SERMs is standard protocol. Triptorelin restarts central hormone signaling while nolvadex blocks peripheral estrogen feedback, creating synergistic recovery. This combination prevents estrogen crash, maintains mood and libido, and preserves muscle mass better than either protocol alone during the critical early post-cycle phase.
